Vos was told not to attack if he came into contact with Jedi, and this is evident in the comic when he’s just running away lol.

this is one of the toughest ones to call, both are quintessential jedi masters, both represent the typical heartiness, durability and natural physiological advantages afforded to their species and therefore enjoy high level energy, endurance, speed and strength over normal humanoids. i would have to give the slight edge to koth.
while agen is a fine, celebrated swordsman- one of the best having mastered djem-so, ataru, makashi as well as unarmed combat, and proves himself one of the light side's chief enforcers, the blunt weapon of the jedi council sent to flatten opposition and apprehend dark fugitives, he is not darth maul-strong, not very muscular, is fairly lean and therefore not as physically imposing or athletically high performing as he could have been. his potential was never completely reached. not just that, his arsenal of force abilities is notably lacking. he knows telekinesis, force valor, augmentation, force assisted jumps and speed and THAT'S IT.
eeth koth is a practiced hand at telekinesis, and has mastered the crucitorn ability to resist torture and pain, on top of his valor, physical augmentation.. so a fairly low force skill base too, but with a trump card there being his rare crucitorn ability. it seems redundant given his species already has a high resistance to pain and mental torture, but the fact he has the ability ON TOP OF his natural abilities, means that his other abilities of natural mental focus and discipline can work in tandem to see him through almost any scenario when he is short handed and has to endure setbacks and injuries. meaning like with grievous torturing him, he can take the pain of electrostaff strikes, while being able to subtly communicate his location non verbally in hologram messages.
eeth koth is also a competent swordsmith in the niman form, the jack of all trades style, and as a jedi guardian, is focused in lightsaber combat primarily and force powers second, just like agen kolar. he is not however as diplomatically bereft as agen kolar, so you could make an argument as to who is the more skilled/useful asset to the jedi order. because while kolar has high praise of his battle skills from mace windu, windu typically compliments his colleagues.. essentially, koth has all kolar's battle skills AND THEN SOME. kolar has been called one of the best duellists in the order, but koth has had a concordance of fealty with windu because their trust was that strong they swapped lightsabers.. kolar has decent victories under his belt and had acquitted himself well on for example geonosis, in his hunt for quinlan and infiltration of the brig- but koth is older, more experienced, has been a council member for about 3 decades and a master for alot longer, has survived his gunship exploding with no others on board surviving save for sora bulq on geonosis (so has pretty much conquered death as far as i'm concerned) is very physically fit and agile and is often sent on high risk frontline missions in wartime because he's dangerous, militaristic and a capable high general and leader, having survived the stark hyperspace war, yiinchorri uprising, jin'ha skirmish, the malastare conflict and later the clone wars. he then survived to face off against vader, putting up a fight for almost 4 minutes before being distracted and stabbed in the back.

Reasons for kolar: stomping vos evdn though vos wasn’t aiming to kill
Reasons for koth: stalemating grievous with a wounded arm before being captured, beginning with like 9 to 1. Also holding his own for over 2 mins against vader in a serious duel.
